Research : Design of a new harmonic multiphoton microscope

Jonathan Belisle got his B.Eng. degree in Engineering Physics at École Polytechnique de Montréal in 2004. He is currently building a Second/Third Harmonic Generation (SHG/THG) Microscope.  For this task, he will be working with a Ti:Sapphire laser that can produce pulses with a duration of 130 fs or less.  The wavelength of these pulses can be tuned by an Optical Parametric Oscillator (OPO) in order to obtain the desired 1.2 µm wavelength.  At this wavelength second and third harmonics are respectively of 600nm and 400nm, i.e. in the visible which makes it easier for detection.  Short laser pulses are necessary in order to have sufficiently high intensity to generate harmonic light while having an average power lower than the sample damage threshold.  He is particularly interested in exploring the possible sensitivity of THG to membrane potential.
